Harley Quinn or formally known as Harleen Frances Quinzel made her first appearance in September 1992 in Batman: the animated series in the episode called the ‘Joker Favour’ with her first appearance many fans fell in love the character, she was voiced by Arleen Sorkin. She was originally was supposed to be a walk on role, but with the feedback from her introduction, she was changed in the Joker famous sidekick and love intrest Harley Quinn. She was known for wearing in a jester outfit in this series. As Tim the artist of the animated series had been friends with Sorkin since college he included parts of her personally into Harley.
Harley then started to make appearance in the DC comics her first appearance in the comic was ‘The batman Adventures issue 12’ keeping the jester outfit.She has been portrayed as a physician psychiatrist and as a psychologist. Then in 1994 in the graphic novel ‘The Batman Adventure:Mad Love’ we were shown the first look of her origin story and people fell in love with the idea as she was known as Arkham Asylums psychologist who fell in love with the Joker as she was treated him, she became an on and off girlfriend for a while before later on changing to Harley Quinn and breaking the Joker out of Arkham. The back story was so good it even won the creators of the graphic novel Best Single Issue Comic of the Year. Due to ‘Mad Love’ creating such a buzz the New Batman Adventure series adapted ‘Mad Love’ in 1999 to an episode which became loved by many fans as they kept it as close to the comic as possible. Thought-out the episode a the series you see Harley been poorly treated by the Joker almost killed by him however she always returned to him as she believed she truly loved him and he loved her. 
After the both Batman series Harley then made several more appearance in the DC universe. You can see her solo potential in the Gotham Girl series as she is one of the four main characters in the series. This series was made to show more of the girl characters in the Batman series. In Gotham girls Harley was apart of the Gotham sirens, in the group was the three most famous girls within DC and they were Catwoman, Poison Ivy and of course Harley Quinn. Throughout Harley Quinn story with the hard time with the Joker, she always turned to Pam or better known as Poison Ivy who despised the Joker not only just for the way he treated Harley. These too were known for being best friends in the series.
Then in 2016 during the ‘Harley Rebirth’ she has a complete look changed in her own comic series. The artist got rid of her famous jester outfit and gave her new twist but keeping it in a jester theme and in this comic series it shows her as a solo villain but with relevant mentions of the Joker making it clear that she still loves him, but while also showing solo endeavours. It linked the look to the fifth series of the suicide squad, making the two look very similar and in the same year of 2016 her look was brought to the life by the actor Margot Robbie who placed Harley Quinn in Suicide Squad.
In August 2014 there was an animated movie made which introduce Harley to the Suicide Squad from the comic book series which include, Killer frost, Dead shot, Captain Boomerang, Killer Shark and KGBeast. This is known as Assault on Arkham and in this movie she was voiced by Hynden Walch. Her look was also related much to the suicide comics series.
In 2009 Harley was then introduced to the batman games series her outfit taking on a completely new look however keeping the red, white and black theme. She was first introduced into Batman:Arkham Asylum. Then in 2011 she made appearance in Batman Arkham city. She was also in Batman: Origins which was released in 2013 and then later on again in 2015 in Batman Arkham knight.
In the 2009 game she was voiced by Arleen Sorkin just like in the original batman adventure series however in the other three games she was voiced by Tara Strong.
She also makes multiple appearance in many batman lego games and movies while also being include in a series on Netflix called DC Superhero girls. Where she is portrayed as a superhero rather than a villain in the series she is paired with her best friend Poison Ivy.
There is also mention of Harley being in a popular series on Netflix known as Gotham which is the backstory to Batman and officer James Gordon. Apparently Barbara Kean is supposed to the Harley Quinn of Gotham as she does from a friendship with Ivy Pepper.
